Determination of the adm (adrenomedullin) gene sequence variation in patients with coronary artery

Abstract (EN)

Coronary artery disease is characterized by atherosclerosis in the vessel wall. In this study, it was intended to determine the relationship between the development of atherosclerosis and the gene sequence variations of ADM known as suppressing the proliferation of the vascular smooth muscle cells which is one of basic mechanisms in atherogenesis.For this purpose, 62 cases with atheroma plaque and 46 controls without plaque were analysed in the study. After DNA isolation from the blood samples of patients with venous for ADM gene sequence variations, ADM gene sequence variations were determined by Sanger sequencing method.A single polymorphism in the promoter (rs3814700 T>C) and 5'UTR (rs545190978 T>G) region of ADM gene, and two different polymorphisms in the exon region (rs5005 C>G (ekson 3)) ve rs767028428 C>T (ekson4)) were found in the present study. In patients, the allel frequencies of the ADM gene polymorphisms were for rs3814700 (T>C) T: %93 and C: %7; rs5005 (C>G) C: %99,2 and G: 0,08; rs545190978 (T>G) T: %99,2 and G: %0,08; rs767028428 (C>T) C: 99,2 and T: 0,08 respectively. It was seen that there was no significant difference between patients and controls in terms of allele frequencies and genotypes of these polymorphism regions determined in the cases. The promoter (rs3814700-C) region polymorphism known as affecting ADM serum level was higher proportion in patients compared with controls. There is a need for studies conducted with serum levels in the large case group to assess the roles of adrenomedullin in the pathogenesis of atherosclerosis.
