Abstract (EN)
In the study presented, the first aim was the synthesis of copolymericstructured hydrogels containing acrylamide (AAm), acrylic acid (AA)and crotonic acid (CA) in different combinations and the studying ofthese hydrogels against changing time, temperature and pH. Theresearchers also aimed to study gels which swell most suitably in thecolon media and the studying of Pantoprazole-Na delivery behaviors.Bio-compatible AAm, AA and CA monomers can easily be polymerizedand provide copolymer. The synthesis of hydrogels by free-radicalpolymerization from aforementioned monomers was realized with thehelp of etylen glycol dimethacrylate (EGDMA) was used as a crosslinkerand with the help of amonium persulfat (APS)/sodium bisulfit as achemically starter. The swelling behavıors of the produced six types hydrogels, againsttime, PAAm, PAAmCA-1, PAAmCA-2, PAA, PAACA-1, PAACA-2 wereexamined in 37,0 ºC during 30 hours. As a result of this studying, it wasvifound out that the swelling values of all hydrogels had increased astime went on and it had reached at a stable value at about 20 hours. The swelling behavıors of hydrogels under the changing heatconditions were watched carefully between 4,0 ºC and 60,0 ºC during 20hours. It was detected that all hydrogels had the lowest swelling valueat 4,0 ºC, the swelling values grow as heat went up and these valueswere stable at about 50,0 ºC.However, the swelling studying of hydrogels against changing pH wereexamined between pH=2,0 and 12,0 at 37 ºC during 20 hours.Consequently, it was found out that the swelling behavıors of PAAmCAhydrogels were a bit influenced by pH change, the swelling behavıorsof PAACA hydrogels first grow with the increasing pH and these valueswere stable at about pH=8,0.For controlled drug delivery studies, the researchers examinedPantoprazole Sodıum (Pantoprazole-Na) delivery at pH=7,4 which wassuıtable for colon environment after choosing PAAmCA-1 and PAACA-1hydrogel. The drug was put into PAAmCA-1 hydrogel by of entrappingand it was also put into PAACA-1 hydrogel via loading. The drugdelivery through PAAmCA-1 and PAACA-1 hydrogels containingPantoprazole-Na via entrapping and loading was decided on UVspectrophotometer and λmax = 288,0 nm.Science code: 201.1.117Keywords : Crotonic acid, Acrylamide, Acrylic acid, Controlledrelease, Pantoprazol sodıum, Hydrogel.Page Number: 97Advises : Prof. Dr. Mehlika PULAT
