Antigenotoxic effects of lipoic acid and ferulic acid against mytomycine-C on human lymphocyte cultures

Abstract (EN)
In this study, antigenotoxic effects of lipoic acid and ferulic acid on MMC induced chromosomal aberrations, sister kromatid exchanges and micronucleus in human lymphocytes was investigated. Preventive efficacy of LA and FA due to their antioxidant properties against the effect of MMC on the mitotic index and replication index was also investigated. In the LA study, all cultures were treated with 0,2 ?g/ml MMC for 48 hours. Three different consantrations (0,5, 1, 2 ?g/ml) of LA was added to the MMC induced cultures for three different times; 1 hour pre-teratment, simultanous treatment and 1 hour post-treatment. In the FA study, the MMC induced cultures were simultaneously treated with five different consentrations (0,25, 0,5, 1, 2, 4 ?g/ml) for 24 hours. A negative control, a positive control and a vehicle treated control (ethanol) was also included. In all the cultures treated with MMC+LA and MMC+FA, the frequency of abnormal cells and CA/cell were reduced compared with MMC treated cells. This reduction was significant in LA treatment. Statistically significant reduction was also observed in SCE/cell and MN frequencies in both LA and FA treatments. These results show anticlastogenic and antimutagenic effects of LA and FA against MMC induced genetic damage. On the other hand, LA and FA treatments induced significant reduction in the mitotic index in all treatments. These results indicate that these these two antioxidants have antimitotic effects in human culture cells.
