Abstract (EN)

Breast cancer (BC) is one of the most common malignant tumors in women and the second leading cause of cancer. Approximately 1.5 million new cases of breast cancer are diagnosed annually, and approximately 460,000 patients die each year from breast cancer chemo-resistance and metastasis. A few breast cancer patients with similar characteristics show different clinical results. Therefore, biological features have limitations regarding diagnosis, prognosis, and prediction of clinical outcomes. New diagnostic and prognostic approaches are urgently needed to identify new personalized treatment modalities that improve the quality of life of breast cancer patients. miRNA profiles have been effectively used to classify breast cancer patients as responding or non-responsive groups. Therefore, it has been clearly demonstrated that miRNAs potentially regulate breast cancer progression and have been used as new diagnostic, prognostic and predictive biomarkers of breast cancer. In this study, after high-level expression of miR638 in MDA-MB-231 breast cancer cell line and miR control lent as a control using viral transduction method, it will be examined whether the cells can incorporate these vectors. To control the expression of miRNAs, the green irradiation induced by GFP in vectors through fluorescent microscope will be taken into account. Proliferation and proliferation ability in cells will be tested using the MTT method. In addition, in order to examine the effect of miR638 in the aforementioned cells, RNA will be isolated from the cells on the second day of transduction and the levels and changes of genes related to apoptosis and proliferation, which are important in cancer cells, will be examined by real-time polymerase chain reaction (GZ-PCR). Finally, we will look at how miR control and miR638 affect migration (migration) with a line drawing method in MDA-MB-231 breast cancer cell line. The aim of this study is to investigate the effects of miR638 on human breast cancer cell lines.
