Abstract (EN)

Nowadays, polymeric drug delivery systems are used extensively. Studies on the diblock and triblock copolymer micelles and shell cross-linked which are a candidate of this group carrier are being continued. Polymeric micelles are important because of their use for drug release.Micelles are usually composed of a hydrofobic core and hydrophilic corona. This structure is an adventage for hydrofobic drugs, active ingredients. At this study, meloxicam, a hyrofobic non-steroidal anti-inflamatuary drug, was used. %0.5 solution of Poly{2-(dimethylamino)methyl methacrylate)}-Poly {2-(diethylamino)ethyl methacrylate)} (DMA-DEA) diblock copolymer and Poly{2-(diethylamino)methyl methacrylate)}-Poly{2-(diisopropylamino)ethyl methacrylate }-Poly{2-(N-morpholino) ethyl methacrylate }(DMA-DPA-MEMA) were used as a polymeric drug carrier system. The polymer was dissolued at pH below 2 by adding acid solution. Forming micelles were occured when this solution was getting basic by adding KOH at pH 8-9 and depending on temperature of the medium. Radius of micelles was determined by using dynamic light scatring (DLS) and zeta sizer. Meloxicam was dissolved in methanol and loaded micelles solution. But methanol damaged to micelles structure so dimethylsulfoxide (DMSO) was used for loading process. Diameter of diblock copolymer? s micelles were increased after loading meloxicam on micelles. This situation shows that meloxicam was locked in micelles. In vivo and ex vivo drug release studies of meloxicam have been done by using Franz diffusion cell and dialysis tubing and the skin from the abdomen of femeale patient.
