Abstract (EN)

The aim of this study was to investigate the effect of environmental factors and CYP1B1 gene polymorphisms on benzo(a)pyrene DNA adduct level in breast cancer patients. A total of 59 female patients aged between 25-60 years participated in the study. Samples were collected between June and December 2022 using sterile disposable medical syringes of 5 mL capacity. Whole blood tests were performed on MS4S and Haematology analyser, BPDE-DNA adduct levels were estimated by ELISA technique and genotypes were estimated by PCR-RFLP technique. For all patients, it was found that there was no significant difference (P>0.05) between Benzo(a)pyrene DNA adduct, White blood cells (WBC), Blood filled cell volume (PCV) and Blood haemoglobin (%HB) parameters and genotypes, while there was a significant association between platelet count (PLT) and CT and TT, TT and CC genotypes. In addition, there was a significant difference in the correlation results of benzo(a)pyrene concentrations for breast cancer patients and whether the patient lived in rural or urban areas. Our study is important in terms of evaluating the health risk when exposed to benzo(a)pyrene levels, as well as utilising the results of the parameters in determining the health status of breast cancer patients and breast cancer development. According to our results, the genotypes of the patients participating in the study were mostly not affected by exposure to polycyclic aromatic hydrocarbons (PAHs). This may be due to human racial differences, but the population environment often influences the incidence of breast cancer depending on the amount of exposure to benzo(a)pyrene levels.
