Investigation of quercetin, alpha lipoic acid and thiazolidinedione administrations on serum resistin levels in metabolic syndrome induced rats

Abstract (EN)

Metabolic syndrome is the most important disease of 20th century.. Insulin resistance component of metabolic syndrome. Resistin, which is recently discovered, come into prominence with its effects on insulin resistance. Unfortunately metabolic syndrome doesn?t have an effective treatment protocol. In this study, determination of serum resistin levels and the effects of alpha lipoic acid, thiazolidinedione and quercetin administrations on serum resistin levels in metabolic syndrome induced rats were aimed to be investigated.A total of 60 Wistar albino rats weighing 250± 20 g were used in the study and rats were divided into 6 groups. Group 1 (n=10, control group; 1 ml/kg normal saline administered i.p. for 10 days); Group 2 (n=10, metabolic syndrome group; 1 ml/kg normal saline administered i.p. for 10 days and metabolic syndrome was induced by adding %10 fructose in tap water for 45 days); Group 3 (n=10, quercetin group; 15 mg/kg/day quercetin administered i.p. for 10 days after metabolic syndrome induced); Group 4 (n=10, ALA group; 100 mg/kg/day ALA administered i.p. for 10 days after metabolic syndrome induced); Group 5 (n=10, quercetin+ALA group; 15 mg/kg/day quercetin and 100 mg/kg/day ALA administered i.p. for 10 days after metabolic syndrome induced); Group 6 (n=10, TZD group; 20 mg/kg/day pioglitazon administered orally for 10 days after metabolic syndrome induced). After all administrations, rats were decapitated and blood samples obtained for biochemical parametersSerum resistin levels were found to be increased significantly in experimentally metabolic syndrome induced rats (% 73,93; p<0.001); and decreased significantly in quercetin (% 35,85; p<0.001), ALA (% 35,69; p<0.001), quercetin+ALA (% 54,15; p<0.001) and TZD (% 29,02; p<0.001) groups. An increase was seen in serum glucose, insulin and HOMA-IR levels in metabolic syndrome group; and it was found that this increase recovered in treatment groups. Similar positive effects seen when lipid paremeters were examined; however the results were better in quercetin, ALA and quercetin+ALA groups compared with TZD group. Especially, more positive effects in lipid profile and uric acid levels in quercetin and quercetin+ALA groups than in TZD group were considerable.In conclusion; there is a statistically significant relationship between metabolic syndrome and serum resistin levels; and the applied treatments affected resistin levels positively. When the treatment groups were compared; we also consider TZD as an available agent in metabolic syndrome treatment. However when cardiac problems and renal damage, which can be developed as a complication of metabolic syndrome, taken into account; we think that quercetin and quercetin+ALA are more effective in treatment.
