Investigation of serum, liver and brain salusins expressions in rats with induced metabolic syndrome

Abstract (EN)
Metabolic syndrome (MetS) is a metabolical disorder which has been getting more and more widespread throughout the world and in our country and although the underlying reasons of the illness have not been totally revealed yet. In this disorder, various factors such as prediabetes, diabetes, abdominal obesity, irregularity in blood fat level and highblood pressure are grouped together. This MetS study has been designed in order to reveal the expressions of hormones salusin-? (Sal-?) and salusin-ß (Sal-ß), which have a role in the regulation of blood pressure and energy homeostasis, in the tissue of the brain and liver vary depending on MetS and how these changes reflect on the amounts of Sal-? and Sal-ß in the circulation. Besides depending on how the concentrations of MetS lipit profile, glucose and uric acid varies. Sprague-Dawley species of 35-40-day 14 male rats, were included in the study. The rats were randomly divided into two groups as control and fructose MetS and put into 4 cages. For three months, the control group was fed with theregular rat food whereas the MetS group was fed with an additional 10% of water with fructose standart as ad libitum. While in serum and tissue (brain and liver) supernatants, Sal-? and Sal-ß concentrations were analysed by ELISA method, the hormonal (Sal-? and Sal-ß) expressions of brain and liver tissues were coloured in a immünohistochemical way.Biochemical parameters were assigned by aoutoanalyser. It was reported that depending on MetS Sal-? and Sal-ß changes declined both in serum and brain and liver tissue superntants. It was shown in a immuhistochemical way that Sal-? and Sal-ß is produced by epitelyum layer in brain and supraoptik nucleus cells in hipotalamus, and hepatosit cells inthe liver. It has been found out that the process of being coloured is less intense in the cells expressing Salusin. There is a statistically considerable increase in MetS groups hunger serum glucose, ALT, GGT, triglyceride, LDL-K, total cholesterol and uric acid levels and a considerable decline in HDL-K levels. According to our results, depending on MetS, the decrease in the salusin expressions in brain and liver lead to their amounts in serum pool. Itis believed that the fall in salusin amounts in the circulation lead to the formation of foam cells and since the blood pressure regulation is disrupted it also results in hypertension, which is one of the criteria of MetS. We believe that through replecement, Salusins? pulling physiological levels up will be effective in getting protected from the harmfuleffects of MetS.Keywords: Metabolic syndrome, Salusin-?, Salusin-ß, immünohistochemistry, liver and brain
