Effect of mesenchymal stem cells on chemically-induced endometrial senescent model

Abstract (EN)
Aim: In this study, it was aimed to evaluate the change of the senescence markers of chemically induced senescent model of human endometrial epithelial cell line (RL95-2) after co-culture with human mesenchymal stem cells (hMSC). Material and Methods: The study was performed at four groups. (i) Control group RL95-2, (ii) HU-induced senesced group RL95-2, (iii) HU-induced senesced before co-cultured with hMSC group RL95-2 and RL95-2 cells that were not senesced but co-cultured with hMSC. After culture period, the cells were stained with X-gal stain for senescence associated ß-galactosidase activity and immunocytochemistry stain were assayed with p16Ink4a and Lamin-B1 antibodies. qRT-PCR was conducted for IL1α and IL-6 -6 expressions. Finally, IL-1α and IL-6 levels on the cell culture media were evaluated by using ELISA. Results: According to findings of the study, levels of p16Ink4a, IL1α and IL-6 were upregulated while Lamin-B1 was downregulated and X-gal positive cell ratio was increased in comparison to control group. However, in the group that were co-cultured with hMSC after HU treatment, levels of p16Ink4a, IL1α and IL-6 were downregulated while Lamin-B1 was upregulated and X-gal positive cell ratio was decreased in comparison to senescent group Conclusion: It can be stated with these findings that mesenchymal stem cells caused decrease of senescence markers on endometrial senescence model. Moreover, they suppressed senescence associated secretory phenotype and showed anti-inflammatory effects. With further studies, new cellular therapies might be developed for the implantation problems which generally is the result of uterine senescence. Keywords: Endometrium, Senescence, Stem Cell
