Abstract (EN)
In this study, preconcentration conditions of Mn(H), Co(II), Fe(ni) and Cr(III) ions for microorganisms immobilized on Amberlite XAD-4 polymer by solid phase extraction method were investigated. Some of the results were compared with the results obtained for Amberlite XAD-4 without any immobilized microorganisms. Saccharomyces carlsbargensis, Escherichia coli and Agrobacterium tumefaciens were studied as microorganisms. The determination of the elements were carried out by flame atomic absorbtion spectrometry. The effect of pH, amount of solid phase, volume and type of eluent, flow rate and volume of sample solution on the recocery of the studied ions were investigated. The recoveries Mn(Il), Co(II), Fe(m) and Cr(m) ions were found as 98±3%, 100±2%, 99±2% and 98±2%, respectively for Saccharomyces carlsbargensis immobilized on Amberlite XAD-4, 98±2%, 99±3%, 99±2% and 9&fc3% for Escherichia coli immobilized on Amberlite XAD-4, %98±3, %99±2, %99±3 ve %98±3 for Agrobacterium tumefaciens immobilized on Amberlite XAD-4, under the optimum conditions determined. The accurracy of the method by using standard reference materials, precision of the method and limit of dedection for the studied ions were determined. The proposed method was applied to water samples, aluminium foils and infant food for the determination of the analytes. Relative standard deviation and relative error were found as about 10% and 7%, respectively for real samples.
