Abstract (EN)

In this thesis study, it is aimed to construct a multi-featured teranostic agent for MR and fluorescence imaging with photodynamic therapy and hyperthermia features. For this purpose, mangan nanoparticles were synthesized by chemical reduction method and conjugation with DTPA bifonctional chelate was provided. Nano-sized non-ionic bilayer surface-active niozome was used as a drug delivery vesicle. Conjugation was done with niozome using the thin layer method. Synthesized Niasome: MnO-DTPA-PPIX NPs will be made effective for photodynamic therapy by surface modification with porphyrin to increase stability of the nanoparticles, decrease cytotoxicity and increase affinity to cancer cells. Using niasome-coated MnO-DTPA conjugation's own fluorescence property, in vitro cell culture studies and bioaffinity tests were performed on PC-3 human prostate cancer cells and BJ healthy skin cells. The dose value to be determined as a result of cytotoxicity studies have been applied for photodynamic therapy. The Niasome: MnO-DTPA-PPIX nanoparticle functionalized with PPIX (protoporphyrin IX) with photosensitizer properties have been examined on BJ and PC-3 cell lines for photodynamic therapy. Photodynamic therapy will be effectively and potentially investigated through the application of phototherapy after radiotherapy first, and then the application of radiotherapy after phototherapy.
