Effects of genotype and plant growth regulators on haploid plant production from anthers of chickpea

Abstract (EN)

In order to determine the effect of auxin type (2,4-D and Picloram) and concentration (5 mg/l, 10 mg/l;1 mg/l, 2 mg/L) as well as cytokine type (BAP and Kin) and concentration ( 0 mg/l, 0.5 mg/l) on haploid plant regeneration from the anthers of the different chickpea genotypes, chickpea cultivars Seçkin, İnci, Onur, Sezgin and Hasanbey genotypes were tested. Results of the variance analyses on the data obtained showed that genotype, auxin type, cytokinin type and concentration as well as their interactions significantly affected the ratio of the anthers forming callus and/or embryo. Ratio of the responding anthers varied between 3.3% and 7.9% depending on the genotype. It was determined that 2,4-D was more efective than Picloram in anther reaction rate. On the other hand, Kinetin resulted in higher anther reaction rate than BAP. When the calli and embryos were transferred into regeneration medium, they regenerated only root but not the plant. From the results of the study, it was concluded that the further research at the aiming to determine response of chickpea genetic resources of Turkey to the anther culture and to optimize the nutrient medium and stress treatment method of the anther must be conducted.
