Abstract (EN)

The World Health Organisation (WHO) defines obesity as an abnormal or excessive accumulation of fat that may impair health. Current data show that overweight and obesity are spreading rapidly as a global epidemic and that 2 billion people, equivalent to approximately 30% of the world's population, have excess body weight. Furthermore, today, the epidemics of obesity and obesity-related diseases have become a major problem for global health and economy. Therefore, it is extremely important to investigate its pathophysiology and treatment methods to combat obesity. In this study, serum asprosine levels and carnitine profile in obese individuals will be analysed to evaluate the potential of these molecules as new diagnostic or therapeutic markers. Asprosin is a newly discovered hormone that is released from fat cells and stimulates the passage of sugar into the blood in the liver. Studies have suggested that asprosine may be one of the causes of many diseases, especially diabetes. Carnitine is a low molecular weight compound obtained from the diet and biosynthesised from the essential amino acids lysine and methionine. Carnitine plays an important role in the mitochondrial oxidation of long-chain fatty acidsCarnitines play a major role in energy metabolism However, the roles of asprosine and carnitine derivatives in obesity are not yet fully understood. Therefore, in this study, the changes of asprosine and carnitine levels in obesity will be determined for the first time and their correlations with body mass index (BMI) will be analysed. In this study, two different obese categories will be formed according to body mass index (BMI) and Blood samples will be taken from 60 obese and 30 healthy individuals, including type-1 (vki=30-35), type-2 (vki=40 and above) and 30 from each group. The amount of asprosine was analysed by ELISA method and carnitine profile was analysed by LC-MS/MS method. The data obtained were evaluated using SPSS and MetaboAnalyst programmes. The correlations of asprosine and carnitine levels with body mass index (BMI) were determined and their potential use in the diagnosis and treatment of obesity was investigated.
