Abstract (EN)

Bean, as an important source of protein, has the leading position among leguminous seeds with 614,948 tons of production. Salinity, which is one of the most important problems in our arable areas, is also affect bean farming negatively. In this study, for the analysis of increasing activity of DREB2A gene by salinity stress application in organic and conventional bean farming while RNA isolation was performing at the same time at 20th hour to stress applied leaf sample genomic DNA isolation was done. PCR was done by designing reverse and forward primers according to DREB2A. PCR products were purified by agarose gel and sent to index analyses by cloning into pGEM-T Easy vector. After plasmid isolation, cutting sites in multiple cloning area were cut by an appropriate enzyme and linearized; then the amount was determined and sent to index analyses. BLASTN and BLASTP analysis were done, to determine the similarities and possible functions between sequences that are recorded in the database directory of the GenBank and the resulting 702 bp long DNA index. As a result of analysis; similarity between cloned gene area and soybean DREB1 gene was observed. To use in Real Time PCR, DREB2A forward and reverse primers were designed from the cloned gene area .Bean actin gene was used as a housekeeping gene. After measuring the purity of the isolated RNA, melting curve analysis were performed for Real Time PCR by synthessing cDNA. Amplification was observed in the melting curve analysis which are done for the bean actin and DREB2A.
