Synthesis of pyridazinone derived compounds with promising antiplatelet effects and investigation of their biological activities

Abstract (EN)

In our study, 26 original compounds have been synthesized including 2- ? 6-phenyl-3(2H)-pyridazinone-2-yl ? etanamide, 2- ? 6-(4-methylphenyl)-3(2H)-pyridazinone-2-yl ? etanamide, 2- ? 6-(4-chlorophenyl)-3(2H)-pyridazinone-2-yl ? etanamide and 2- ? 6-(4-nitrophenyl)-3(2H)-pyridazinone-2-yl ? etanamide derivatives and ethyl 2-(6-(4-nitrophenyl)-pyridazine-3(2H)-one-2-yl)acetate and 2-(6-(4-nitrophenyl)-pyridazine-3(2H)-one-2-yl)acetic acide derivatives. The chemical structures of the compunds are shown in Scheme-5.The structures of the synthesized compounds are elucidated by elemental analyses and their IR, 1H-NMR spectral data.The antiplatelet effect of the compounds were screened by turbidimetric based agregometry method using aspirin as reference. The compounds considered to have antiplatelet effects according to the the literature data, were found inactive in our experimental studies, and this was incorrelation with the PASS program.According to the data originated form the PASS program, the compounds, lacking to have antiplatelet effects in biological tests, were found convenient to make research and development studies for the use of multiple-sclerosis therapies.
