Abstract (EN)

This study focused on the characterization of Cyanobacteria founded in Porsuk River and Seydisuyu.The sampling was performed during year 2006 from 21 different stations in the Porsuk River and 5 different stations in the Seydisuyu and boron waste pool of Etibor Establishment. 141 Cyanobacterial isolates were obtained with BG-11 medium. In addition to microscobic analysis, molecular analysis also was applied for identification of Cyanobacterial isolates by using 16S rRNA PCR amplification of genomic DNA.Water samples collected from Porsuk River, Seydisuyu and boron waste pool at 2006 October were analysed by using culture independent methods such as denaturating gradient gel electrophoresis (DGGE) and 16S clone library. Cyanobacterial composition found in Porsuk River and Seydisuyu was shown for the first time by using microscopic and molecular studies.With the cultural methods, strains belong to Synechococcus, Cyanobium, Anabaenopsis, Leptolyngbya, Microcoleus, Pseudanabaena, Synechocystis, Oscillatoria, Snowella, Nostoc, Phormidium, and Limnothrix genera in Porsuk River and Pseudanabaena, Anabaenopsis, Leptolyngbya, Phormidium, Synechocystis and Nostoc genera in Seydisuyu were obtained. The isolates belong to only Leptolyngbya genus was obtained from boron waste pool.Denaturing gradient gel electrophoresis (DGGE) fingerprint analysis based on BLAST results of 16S rRNA gene sequences based on the revealed that Porsuk River water sample may have species from Arthrospira, Lyngbya, Planktothricoides, Microcystis genera, Seydisuyu water sample have Cyanobacterium, Lyngbya, Nostoc, Leptolyngbya and Phormidium genera where the boron pool water samples have representative species from Oscillatoria, Nostoc, Phormidium, Arthrospira, Planktothricoides, Aphanothece, and Leptolyngbya genera. With the cloning method Leptolyngbya, Merismopedia, Nostoc genera were obtained from boron pool. Phylogenetic and BLAST analysis of some representative partial sequences indicates that most of the 16S rRNA sequences were under currently unavailable nucleotide sequence databases. As a result, we conclude that novel Cyanobacterial species may be present in Seydisuyu and boron pool.
