To investigate relationship between polymorphisms of the ELAC2 and the SRD5A2 genes and prostate cancer

Abstract (EN)

Prostate cancer can be determined as the alteration of the balance between cell proliferation and cell death in the prostate bladder which causes malign increase of the organ volume. The factors causing prostate cancer can be hormonal, diet, environmental and genetical. There are a number of genes causing prostate cancer. These are ELAC2, SRD5A2, HPC1, HPCX, AR, PSA etc. genes. ELAC2 gene composed of 24 exons is 25.1 kb and has been mapped to chromosome 17p11.2. Steroide 5?-reductase type II enzyme has a 28.398 kDa molecular weight. The responsible gene is called as SRD5A2 that is consisted of 5 exons and 4 introns. This gene has been mapped to chromosome 2p22-23 and has approximately 56.4 kb length. The aim of this study is to investigate relationship between Ser217Leu and Ala541Thr polymorphisms of the ELAC2 gene and Ala49Thr and Val89Leu polymorphisms of the SRD5A2 gene and prostate cancers. For this aim, regions where polymorphisms are located in were amplified with Polymerase Chain Reaction (PCR) method. Polymorphisms were determined by using available restriction enzymes with Restriction Fragment Length Polymorphism (RFLP) method. In this study, a statistically significant relationship has been found between ELAC2 gene Ser217Leu (p=0,001<0,05) and SRD5A2 gene Ala49Thr (p=0,001<0,05) polymorphisms and prostate cancers. However, the relationships between Ala541Thr (p=0,494>0,05) polymorphisms of ELAC2 gene and Val89Leu (p=0,460>0,05) polymorphisms of SRD5A2 gene and prostate cancers hasn?t been found statistically significant.Key Words: Prostate cancer, ELAC2 gene, SRD5A2 gene
