Investigation of the effect of noise on oxidative stress and prestin level and the effectiveness of Ginkgo Biloba

Abstract (EN)

In this study, the effects of noise on oxidative stress and prestin levels were investigated, and the effectiveness of Ginkgo Biloba on the changes that occurred was evaluated. In the study, 28 pieces female Wistar-Albino rats aged 2.5 - 3 months, 7 in each group, were used. The 1st Group is Control, 2nd Group is Noise group, 3rd Group is Gingko Biloba group and 4th Group is Noise + Gingko Biloba group. The groups exposed to noise were subjected to noise with a frequency of 8000 hertz and an intensity of 100 dB for 12 hours from 7 pm to 7 am for 21 days. Ginkgo Biloba was administered to the groups receiving Gingko Biloba at a dose of 100 mg/kg/day immediately after exposure to noise. At the end of 21 days, blood samples were taken and examined. In the plasma obtained after centrifugation of blood samples, prestin level was analyzed by Rat-Prestin specific ELISA kit method and for oxidative stress; MDA level which is an indicator of lipid peroxidation and GSH, GSH-Px and CAT enzymes were measured and statistically evaluated. Rats exposed to noise had a one significant increase in prestin levels. Therefore, although prestin is not an alternative to audiogram, it is thought that it be used as an auxiliary biomarker in associating noise with prestin level. In addition, increased MDA level and decreased CAT activities indicate that oxidative stress occurs. It has been observed that the application of Ginkgo Biloba, which has been audiologically tested many times and has antioxidant properties, reduces the increased prestin level due to noise and oxidative stress by removing free radicals.
