Abstract (EN)
The flourishing world population fighting against cancer has caused to focus on novel strategies such as the discovery of novel therapeutic antibodies and the production of biosimilars for cancer therapy. The aim of this thesis is the development of plasmid vectors containing the humanized anti-HER2 antibody gene for the development of the mammalian cell line producing the monoclonal antibody Herceptin (Trastuzumab®, Roche) biosimilar is targeting the HER2 extracellular domain and can be used in the treatment of HER2 overexpressing breast cancer patients. The development of mammalian expression vectors was carried out by gene optimization, cloning and transformation and was verified by restriction enzyme digestion, multiplex PCR and DNA sequencing. In the master's thesis, the pIRES2/LC/HC containing both the light and heavy chains of the anti-HER2 antibody, pIRES2-EGFP/LC containing only the light chain of the antibody and the pIRES2/HC containing only the heavy chain of the antibody were obtained. In additon, the obtained vectors were produced in E.coli host cells.
