Investigation Of The Frequency Of Crp Gene Polymorphism In Ra And The Relationship Between Clinical Symptoms Of Ra And Crp Gene Polymorphism In Rheumatoid Arthritis Object: Serum CRP level is widely used to evaluate disease activity in patients with rheumatoid arthritis (RA), and is also used to calculate disease activity score (DAS28). Differences in genotype distributions of CRP have significant effects on RA pathogenesis. And these genotype distrubitions play important role at management of disease. CRP levels are under genetic influence and single nucleotide polymorphisms in the CRP gene and haplotypes have been associated with CRP levels in active inflammation. Our study is the first study to evaluate the CRP gene polymorphism in patients with RA in Turkey. In our study, we investigated the frequency of rs1205, rs1130864, rs1800947 single nucleotide polymorphisms identified in CRP gene in RA patients and healthy controls. We also aimed to investigate the relationship between CRP gene polymorphisms and serum CRP level and DAS28 score in the RA group. Material and Method: We included 120 randomly selected rheumatoid arthritis patients who met the diagnostic criteria of rheumatoid arthritis. We formed a control group consisting of 100 healthy volunteers who were compatible with the patient group in terms of age and gender, without any health problems and no signs and history of RA. DNA samples were genotyped for rs1205, rs1130864 and rs1800947 using the Taq Man system. Results: In terms of the frequency of CRP gene polymorphisms, the ratio of TT Homozygote and CT Heterozygote was significantly lower in the rs1130864 polymorphism patient group compared to the healthy control group (p: 0.002). There was no statistically significant difference between rs1205 polymorphism and rs1800947 polymorphism and study groups. The rate of TT homozygous cases of rs1130864 polymorphism was higher and CT heterozygous and CC homozygous individuals were lower in patients with lung involvement (p: 0.017). No significant correlation was found between CRP polymorphisms and serum CRP level and DAS28 score in active disease. No statistically significant relationship was found between other clinical and laboratory findings of RA and polymorphisms. Conclusion: We found that CRP polymorphisms had no effect on CRP level and DAS28 score in RA patients. There was a significant relationship between rs1130864 TT homozygous polymorphism in patients with pulmonary involvement. To achieve more accurate results, larger patient groups need to be studied.
