Abstract (EN)
Addictive substrates had been used for both therapy and pleasure in prehistoricalages. As a therapeutic agent, people use addictives as pain relievers. One of themost important health problem is to use of addictive substrates all around theworld in 21st century, particularly those involve in opioid group, and relatedhealth complications.This study investigates effects of deprivation syndrome caused by an opioidantagonist nalokson on oxidative stress, enzymatic and non enzymatic systems.Heroin dependency is achieved by fixed (1 mg/kg) and increasing dose (0.01mg/kg, 0.1 mg/kg, 1 mg/kg and 10 mg/kg) administration of heroin to albinoSprague-Dawley. Nalokson (0.1 mg/kg) is applied the groups in deprivationsyndrome. The animals that are in deprivation syndrome are not treated whileanother group is treated by melatonin (10 mg/kg). Vitamin E + Se (30 mg/kg +100 ?g/kg) is also applied to the third group. Oxidative stress and antioxidantsystems are measured in blood in these following groups: The control group withno addiction and no therapy, the group only with heroin administrated and thegroup in deprivation syndrome as a result of nalokson administration.This study aims to investigate the effects of oxidative stress level in deprivationsyndrome occurred with nalokson and antioxidant therapy ( melatonin, vitamin E+ Se) on rats addicted by giving fixed and increasing dose of heroin.MDA levels are found higher according to SOR amounts on the rats which aregiven heroin and nalokson. It is observed that while GSH levels which are one ofthe important antioxidant decreases, GPx level increases.It is also found that MDA levels decrease and GSH, GPx levels increase withmelatonin and vitamin E + Se therapy after giving nalokson on dependent rats.This shows that antioxidant therapy reduces LPO and causes to increase the levelsof antioxidant by supporting defense system.Because of the toxic effect formed by heroin, oxidant-antioxidant balance ofmetabolism is damaged. SOR amounts increase due to heroin. This studyconcludes that the oxidative stress, which is occurred by the effect of SOR, couldbe decreased with vitamin E + Se and melatonin therapy.
