Abstract (EN)

In this study, isolastion of prokaryotics mircoorganisms from sediment samples taken 4 different location with varied depths of Sakarya Riverhead located in Çifteler town of Eskişehir province was performed. Based on 16S rRNA gene region's nucleotide sequence analysis data of isolates was obtained and phylogenetic analyses was caried out for molecular typing. Sediment samples was collected from river by sterile jar. Sediment samples was applied Dilution plate technique and samples were inoculated on 5 different media including the antibiotics previously prepared. PCR studies of 16S rRNA gene region of 22 organisms isolated from 5 different media was completed. According to nucleotide sequence analysis data of 16S rRNA sequencing; isolates belonging to Cellulomonas, Micromonospora, Saccharomonospora, Brevibacillus, Paenibacillus and Microvirga genus were determined. Phylogenetic analyses with the closest type of 22 isolates were performed via the Mega 7 package program. Similarity percentage and nucleotide difference of isolates compared to the closest type strains were established by using BIOEDIT program. The probability of new species among the isolates were determined. According to phylogenetic analysis of 16S rRNA gene region; isolates S1S32 (Microvirga sp.), S2S24 (Cellulomonas sp.), S1I41 (Brevibacillus sp.), S1S22 (Paenibacillus sp.), S2S21 (Saccharomonospora sp.), S1S33 (Micromonospora sp.) and S1G21 (Micromonospora sp.) could be new species.
