Investigating the polymorphism of TLR2 gene and its association with somatic cell count i̇n the milk of aleppo goats raised i̇n Şanlıurfa province

The aim of this thesis was to investigate the polymorphism in Toll-Like-Receptor 2 (TLR2) gene in Aleppo goats raised in Şanlıurfa province and to determine whether there is a relationship between the detected polymorphism and somatic cell count in the milk obtained from this goat breed. In Şanlıurfa province, 50 ml milk samples were collected from 100 Aleppo goats in 5 farms in Aleppo goat breeding in Şanlıurfa province twice at one-month intervals after the births started. Twenty-five ml of the milk samples were used for DNA isolation and the other 25 ml were used for somatic cell count determination. After 442 bc of TLR2 gene was amplified by PCR, PCR products were treated with VspI enzyme. Three genotypes as GG, GA and AA were observed by analyzing the cut products in 2% agarose gel electrophoresis. It was observed that 95 of the genotyped samples had GG, four had GA and one had AA genotype, the frequency of the G allele was 0.97 and the frequency of the A allele was 0.03. The difference between the genotype averages in terms of the actual somati ccell count in goat milk was statistically insignificant, but after logarithmic correction, the statistical difference was found to be significant and the number of somatic cells in the milk of individuals carrying the A allelewaslower. G/A polymorphism of TLR2 gene wasfoundtoave a significanteffect on SCC. If the relationship between this polymorphism and somatic cell count is determined in thisbreed, this polymorphism can be used as a marker to be used in selection for breeder selection for breeding studies in terms of somatic cell count and mastitis resistance.
