Abstract (EN)

In this study, a gel of hydrophobic interaction chromatography for purification of Paraoxonase (PON1) enzyme, which are important physiological function in metabolism with detoxification, antioxidant activity and related with cardiovascular diseases were synthesized. The hydrophobic interaction chromatography gel was synthesized with Sepharose-4B-L-tyrosine and 1-naftilamin as hydrophobic ligand. Sepharose-4B was activated with CNBr and than L-tyrosine was added as extension arm. Human serum paraoxonase (PON1) was purified from waste blood samples, which taken from Siirt health organization services, by ammonium sulfate precipitation and synthesized hydrophobic interaction chromatographic gel. The in vitro effects of different concentrations of Be, Mg, Ca, Sr and Ba which doped on cerium vanadate (CeVO₄) derivatives, on purified Paraoxonase (PON1) were researched. The structure of %10 Mg CeVO4 (%10 Mg- CeVO4) in the mentioned cerium vanadate derivatives was determined as the best inhibitory effect on PON1 with 1.1710-4 M concentration.
