Abstract (EN)

Acrylamide is a toxic chemical which is widely used in industry. Acrylamide could occur in food during high temperature cooking and it can be taken up by humans via diet. Adverse effects of acrylamide on human body are completely unknown when taken up via food chain. Therefore, dietary source of acrylamide could pose a great risk for public health. Acrylamide, which in food, is classified in Group 2A as a `possible carcinogen to humans? by International Agency for Research on Cancer (IARC).In this study, it was aimed to investigate the possible toxic effects that subchronic doses of acrylamide with administered drinking water on the liver tissue and to test these effects preventability with organic dried apricots in female Spraque Dawley rats.Forty female Spraque Dawley rats with the average weight of 200 g were divided into 4 equal groups as follows: Group 1 (control); animals in this group was administered with normal diet and normal drinking water for 12 weeks. Group 2 (apricot); animals were administered daily diet with %5 apricot and normal drinking water for 12 weeks. Group 3 (acrylamide); animals were administered daily acrylamide at 500µg/kg via drinking water and normal diet. Group 4 (acrylamide-apricot); animals were administered daily acrylamide 500µg/kg via drinking water and diet with %5 apricot. At the end of 12 weeks-period, all rats were decapitated. Samples of liver were collected for histopathological examination as well as biochemical analyses to measure levels of Glutathione Peroxidase (GSH-Px) aktivity and Glutathione S-Transferase (GST) activity, Glutathione S-Transferase Pi gene expression, Malondialdehyde (MDA), Reduced Glutathione (GSH) and Total Protein. As a result of our study, liver tissue MDA levels of acrylamide group were significantly lower than apricot and apricot-ACR groups, GSH levels, lower than the control group, GSH-Px levels were lower than control and apricot groups. Apricot group liver tissue MDA levels were significantly lower compared to the acrylamide group, GSH levels were lower than the control group, GSH-Px levels were found higher compared to the group of acrylamide. GST and GST-Pi gene expession levels were no significant differences between the groups. All the results obtained, the rats daily for 3 months 500 ? g/kg dose of acrylamide in the liver tissue without any histopathological damage has been detoxified by the liver easily. Did not bring serious damage to the liver tissue of acrylamide in a histopathological and biochemical parameters that have not signed because of a serious change in the way, for a period of 3 months 5% of the organic dried apricots with diet a protective effect on the liver and the matter is not fully exposed. Therefore, the protective effect of apricot against toxicity from acrylamide, the examination of studies must be done by applying higher doses of acrylamide.
