Optimization of rabbit liver arginase and distribution of arginase in tissues of rabbit

Abstract (EN)

2. ABSTRACTIn this study, for measuring of arginase in rabbit tissues was aimed to optimize of arginase in liver tissue. Optimum pH, preincubation temperature, preincubation time, incubation time, Mn++ concentration, and L-arginine concentration of arginase in liver tissue was determined. Arginase levels were measured in different tissues of rabbits after conditions were optimized.Its was found to be preincubation temperature 65 °C, preincubation period 13 minutes, incubation period 10 minutes and optimum pH 10 for arginase in rabbit liver tissue. Enzyme achieved its highest activity at 2 mM MnCl2 concentration. Consequently, it was determined that Mn+2 ions and preincubation at 65 °C were essential for the activation of the enzyme. Km of arginase in rabbit liver tissue for L-arginine were measured to be 1,6 mM.Levels of the arginase enzyme in liver, kidney, heart, uterus, brain, spleen, intestine, esophagus, muscle, tongue, lung, stomach, trachea tissues of rabbits were determined and found as 105,09 ± 12,91; 27,03 ± 3,37; 7,55 ± 0,91; 3,03 ± 0,23; 3,22 ± 0,60; 2,00 ± 0,21; 1,91 ± 0,07; 179 ± 0,44; 1,76 ± 009; 1,60 ± 0,64; 1,56 ± 0,18; 1,56 ± 0,18; 1,08 ± 0,57; 0,45 ± 0,07 unites, respectively. Among the studied tissues, the highest arginase activity was found in liver, the lowest arginase activity was found in trachea.Key Words: Rabbits, Liver, Arginase
