Abstract (EN)

Aim: In our study, it was aimed to investigate the role of bone marrow-derived mesenchymal stem cells (BMDSC) in the model of Asherman's syndrome brought to the rats, removal of adhesion from the center and the role of blastocyst in endometrium implantation and effects on the molecules involved in this process. Materials and Method: The Asherman model is chemically formed. Therapy; four different groups were set up to be provided nurtient (BY), bone marrow-derived mesenchymal stem cells (BMDSC), and 48-hour nutrient (Nish). First group (G1); Asherman + BY, second group (G2); Asherman + Niche, third group (G3); Asherman + BMDSC and fourth group (G4); Asherman + BMDSC + Niche. Each group was divided into three subgroups and histochemically staining were performed hematoxylin-eosin and masson trichrome and immunohistochemical staining such as TGF-β1, PCNA, eNOS, VEGF, Stro-1, c-Kit, Laminin, Fibronectin and L-Selectin. Also TUNEL staining was performed for apoptosis. Results: Statistically significant difference (p <0,05) was found between the groups in terms of both histologic and neonatal morphologic evaluations in BMDSC and BMDSC + Niche groups compared to the other groups. It was found that BMDSC and Niche of could remove the adhesions in uterine adhesions, making the endometrium more efficient for implantation. Conclusion: The therapeutic properties of BMDSC's in endometrial injuries, increased thickness of the endometrium, number of glands and vascularization, reduction of fibrous areas, implantation of embryos in the adhesive areas, and increased number of newborns are promising for the transfer of experimental Asherman studies to the clinic. It has been shown that BMDSC and Niche applications may contribute to clinic through treatment of adhesion molecules in the mechanisms causing infertility.
