Synthesis and structural characterization of ß-diketonate based ligand and its lanthanide chelate to be used in preparation of medical diagnosis kits

In this study we aimed to synthesize β-diketonate based ligand and its Eu(III) complex which used in medical diagnosıs kits. For this purpose, we chose acetophenone as the starting material which contains methyl group at the alpha position of the ketone group, after literature research results. Firstly, we obtained 4,4,5,5,6,6,7,7-octafluoro-1,10- diphenyldecane-1,3,8,10-tetraone (H2ODİT) ligand between the reaction of acetophenone and diethyloctafluoroadipate with claisen condensation. This ligand was charactrezied by 1H-NMR, 13C-NMR and mass spektrum analyse. We were obtain single crystal of H2ODİT ligand and characterized by X-ray analysis. The X-ray analysis shows that this ligand has keto-enol tautomerism. At the second step ın order to bind H2ODİT to the antibody, it is reacted with chlorosulfonic acid. As a result of this reaction the functional group of -ClSO2 was bound. This group was bound carbon atom at the ortho position depending of carbon atom which bound keto group of acetophenone in H2ODİT molecule. The structure of H2CODİT was characterized by NMR and mass spektrum analysis. In the third step, to understand the usability of the ligand as a biolabel, the complex were synthesized with EuCl3. After dried and purified the complex compound the luminescence analysis was performed. When the complex was excited with UV light at 306 nm wave length, specific hypersensitive 5D0 → 7F2 phosphorescence transition of the Eu (III) observed as the literature indicatig at 615 nm. Proven that, it was understood the H2CODİT molecule can work as biolabel to be used in medical diagnosıs kits.
