Abstract (EN)
Oxidative events occurs during wound repair process just as in normal metabolic process. Platelet derived growth factor (PDGF) is one of the important growth factors that play a role in wound healing. In the present study, we aimed to investigate time-dependent effects of exogenous PDGF administration on oxidative events during dorsolateral cutaneous wound healing in rats.Experiments were performed on 42 female Wistar albino rats (weight: 200-250g). Animals were divided into four group: control group (n=6), untreated group (n=12), chitosan-treated group (n=12) and chitosan+PDGF-treated group (n=12). Two full-thickness excisional skin wounds were made under anesthesia in all animals except for control group. In the chitosan+PDGF-treated groups, the wounds were treated topically with a single daily dose PDGF-BB (7 ng/ml) after wounding. In the chitosan-treated groups, the wounds were treated topically with equal amount blank chitosan gel. After these administrations, on the 3th and 7th days of wound healing, the animals were sacrificed. Thiobarbituric acid reactive substances (TBARS), nitric oxide (NOx), ascorbic acid (AA), glutathione (GSH) levels and superoxide dismutase (SOD) activity were measured spectrophotometrically in wound tissues. Results were expressed as mean ±standard deviation. Mean differences were compared by Anova variance analysis (P<0,05).PDGF significantly increased TBARS levels in early phase of wound healing. In contrast, it significantly decreased TBARS levels in later phase of healing. In the chitosan+PDGF-treated group, NOx levels were decreased on day 3 and 7 when compared with the chitosan-treated groups. Non-enzymatic antioxidant levels increased by PDGF administration and this may have contributed to increase of wound tissue antioxidant capacity. In light of this information, PDGF administration may have altering effects on oxidative balance depending on time in wound healing process.Key Words : PDGF, Wound Healing, Oxidative Stress, Antioxidant, AA, NOx, GSH
