Abstract (EN)
Gynogenic plants were produced from Allium ampeloprasum genotypes and the conditions affecting them were determined. It was found that the most suitable media for ginogenesis stimulation in this species were BDSD (2,4-D and 100 g/l sucrose), which produced 0.22% ginogenic plants, and BDSA, which produced 0.21% ginogenic plants, while the most suitable media for somatic shoot stimulation was BDSC, which produced 5.50% somatic shoots. These results indicate that media and genotype have strong effects on ginogenesis induction and somatic shoot induction in A. ampeloprasum species. As a result of the ginogenic analyses of these plants, it was shown that A. ampeloprasum plants are mostly diploid and some of them are tetraploid and methods to investigate this genetic feature are discussed. All materials in this thesis responded to ginogenesis and somatic shoot stimulation. The data obtained show that it is possible to realise the desired breeding and clonal propagation applications in edible Allium species at the in vitro level. In this aspect of this thesis study, the ginogenic production of plants related to edible Allium biotechnology in our country has been demonstrated and their genetic study and related methods have been discussed in terms of planning future studies. The importance of producing economically important plant species in our country with similar methods and their genetic and bioinformatic analysis was emphasised.
