Abstract (EN)

Aim: It was aimed to investigate the microorganisms causing bacterial vaginosis (BV) and vaginitis in women admitted with vaginal discharge by culture, microscopy and molecular methods. Materials and Methods: Three sterile swab swab samples taken from 100 female patients aged 18-50 years who applied to Dicle University Medical Faculty (DÜTF) Department of Obstetrics and Gynecology with vaginal discharge between July 04, 2018 - August 30, 2018 were included in the study. Two swab specimens were used for Gram staining and culture processes. The Gram stained smears were investigated for the presence of polymorphonuclear leukocytes (PNL), Clue cells and bacteria with different morphologies. The isolates grown on culture were identified up to genus or species with MALDI TOF-MS. The other swab was used to investigate the agents that were difficult to grow in culture by multiplex-polymerase chain reaction (PCR). Results: A total of 100 female patients with vaginal discharge were included in the study. At least one microbial agent was found in 63 of these patient samples. All smears were evaluated according to Nugent score; 27 of them as positive for BV, 39 smears had intermediate scores and 34 smears were found negative for BV. A total of 29 (29%) Candida spp., 9 (9%) S. agalactiae, 3 (3%) S. aureus, 2(%2) S. anginosus, 1(%1) G. vaginalis, 1(%1) S. disgalactia and 1(%1) S. haemolyticus were isolated. DNAs of 23 (23%) M. hominis, 13 (13%) U. urealyticum, 9 (9%) T. vaginalis and 3 (3%) C. trochomatis were detected by multiplex-PCR method. In Conclusion: Candida spp.were the most frequently isolated agents by culture method, in our study. M. hominis, U. urealyticum, T. vaginalis and C. trachomatis DNAs were detected by multiplex PCR method in 23, 13, 9 and 3 samples, respectively. Gram staining and culture methods alone are often not enough for the diagnosis of patients with vaginal discharge. Molecular diagnostic methods provide an important contribution to the detection of vaginosis and vaginitis. Keywords: Bacterial Vaginosis, Vaginitis, Candida spp., Multiplex PCR
