Abstract (EN)
Renal dysfunctions are increasingly seen recently. Today, the negative effect of obesity on the kidneys is increasingly attracting attention. Since treatment of kidney dysfunction requires intensive labor and financial burden, including dialysis therapies, necessary precautions should be taken in the risk group before kidney dysfunction develops. Chromium (Cr) is a trace element and plays an important role in the regulation of lipid and carbohydrate metabolism. It may therefore be useful in preventing kidney damage associated with obesity. The aim of this study is to investigate the possible effects of chromium histidinate (CrHis), which is the organic form of chromium, on kidney function in rats fed high-fat diets and to investigate the effects of SGLT-2, NOX4, L-FABP, HO1, Nrf2, KIM1 and NfkB protein levels in the kidney. In this study, 42 Sprague-Dawley rats were randomly divided into six groups (n =7). Rats 1: fed a standard diet (Control); 2: Feeding on a High Fat Diet (YYD) 3: CrHis purified isomer 1 (CrHis1) with YYD; 4: Isomer 2 (CrHis2) purified by CrHis with YYD; 5: CrHis-purified isomer 3 (CrHis3); 6: YYD and three isomers were grouped as fed with a mixture of CrHis (CrHisM). Although CrHis isomers are from different sources, rats were given the same amount of Cr (10 μg elemental Cr / kg CA / day). At the end of the experiment, the kidney tissues of the rats were taken for analysis. Sodium glucose transporter 2 (SGLT-2), NADPH oxidase (NOX4), Liver-type fatty acid-binding (L-FABP), Hemeoxygenase1 (HO1), Nuclear factor erythroid 2 associated factor 2 (Nrf2) and Nuclear factor kappa B (NfkB) levels of these proteins are Western blot and Kidney Damage Molecule-1 (KIM- 1) It was detected with Eliza. While there was an increase in L-FABP, NFĸB, NOX-4 proteins and KIM-1 levels in kidney tissues of rats fed with HFD compared to control groups; Decreased protein levels of HO-1, Nrf2 and SGLT-2 were observed. While a decrease was observed in L-FABP, NFĸB, NOX-4 and KIM-1 levels compared to YYD groups in rats administered CrHis forms; Increases in HO-1, Nrf2 and SGLT-2 levels were observed. In conclusion, the supplementation of CrHis is protective against renal injury, through supression of oxidative stress and inflammation in rats fed high fat diet.
