Abstract (EN)

The aim of this study is to investigate the day-night differences of locomotor activities, the effects of the memantine and the diurnal rythm of this effects if there is any at Huntington's disease model.The drugs used on this study applied on six different experimental groups that are created randomly and applied half of the groups at daytime (0900), half at night (2100). %0.9 NaCl solution injected to control group of mouse as i.p. An intoxication protocole picked for 5 days to create Huntington model and 3NP (40-120mg/kg) been applied increasingly. At the end of the period of intoxication, cumulative 3-NP dose is 360 mg/kg (at 3-NP groups). Hind limb and truncal dystonia shows that Huntington's disease is created by 3-NP injection. 20mg/kg dosed memantine applied 30 minutes after 3-NP injection to research the effects of memantin i.p. on process of intoxication protocole.Groups are tested on rotarod at the begining of intoxication, on days 1,6 and 9 and the time to ability to stay on rotarod been recorded to estimate the locomotior activities.The application of 3-NP at night or daytime doesn?t have any effect at Huntington model as locomotor activity. The application of memantine effected the performance of mouse at rotarod, as decreasing the time to be able to stay, also effects didn?t show any diurnal rhythm with the time of application.These findings shows that at Huntington's disease model created by 3-NP doesn?t have day-night differences as locomotor activities and memantine effects badly the locomotor activity created by 3-NP.Key Words : 3-nitropropionic acid, Huntington?s disease, memantine, choronobiology.
