Dr. Mehmet ÖZASLAN Doç. Dr. Mehmet GÜLDÜR The production of grapevine is important agricultural sector in Turkey. The total grapevine area is 565.000 hectare and the mean production is 6.46 ton/ha in Turkey. But, the production is lower than the other Mediterranean countries. The main reason of low yield is the effects of virus diseases on the host especially grapevine fanleaf virus (GFLV) in our region. It has been the most important virus among them and transmitted by graft and vector nematodes. There is not enough detailed research on the vector nematode and virus relation in Turkey. In this study, determination of GFLV and their occurrence areas were studied by serological and molecular techniques in grapevine and vector nematodes in Adana and Mersin Provinces. It is found that 63 (16.4 %) samples from total 384 grapevine samples collected from vineyards were infected with virus by DAS- ELISA method. The occurrence areas of vector nematodes and their population densities in the GFLV infected vineyards were detected. Of 307 soil samples collected from the rhizosphere of grapevine, 66 (21.5 %) and 275 (89.6 %) were positive for Xiphinema index Thorne et Allen and X. pachtaicum Tulaganov, respectively. GFLV is found only in X. index by DAS-ELIS A and RT-PCR. In addition to this, GFLV is transmitted only to Nicotiana benthamiana Domin. among lots of weedy indicator plants by mechanical transmission.
