The isolation, typing, antimicrobial susceptibility and detecting of some virulence genes by pcr of Pasteurella multocida strains in cattle in region of Afyonkarahisar

In this study, 650 samples have been collected for the isolation and typing of P. multocida from slaughterhouse and cattle farms in the province of Afyonkarahisar. 300 lung samples from slaughterhouses, 200 swap (healthy 170, unhealthy 30), and 150 tracheal lavage (healthy 130, unhealthy 20) from farms were obtained.In the study, 48 (7.4%) P. multocida strain were identified in all of 650 samples of cattles by phenotypic, serological and PCR method. Genotypic and serological methods were used in the determination of the capsular groups. Hyperimmune sera from New Zealand rabbits immunized with standard P.multocida strain were prepared. In the study isolated 48 P. multocida strains with Slayt Agglutination test 46 (95.8%) were found as serogroup capsular A and 2 strains were untyped. Also these strain were confirmed using by primers specific to KMT1 and OMP genes by PCR. Using specific primers of the capsular group (Townsend et al., 2001) 48 strain were confirmed as in Carter A (100%) capsular group by PCR .In this study, isolates of 48 (7.4%) P. multocida strains were determined by antibiotic susceptibility test. Of the strains 95.8% florfenicol, 93.8% ceftiofur, 91.7% amoxicillin-clavulanic acid were observed to be sensitive. Resistance rates were 22.9% oksitetracycline, 20.8% peniciline G and erythtomicine.
