Detection and characterization of Cucumber Mosaic Virus, (CMV) in the greenhouse pepper cultuvation in Mediterranean region

Abstract (EN)
This study was conducted to detect and characterize Cucumber Mosaic Virus (CMV) in the greenhouse pepper cultivation areas of Adana, Mersin, Hatay and Antalya provinces between 2016-2018 years. During on field observation, totally 300 samples including leaf, shoot, fruit, seed from 263 plants which are suspected as symptomatologic infected with CMV were collected and were tested by DAS-ELISA. Of the 300 tested samples, 62 were found to be infected with CMV, and 23 samples were considered to be diseased with a low absorbance value. RT-PCR studies were performed with 16 isolates from a total of 85 samples identified as positive for DAS-ELISA using a primer pair specific for CMV-specific RW8 / RV11 and a band size of 650 bp was obtained. Eight isolates were selected and DNA sequencing was performed to represent the survey areas. PCR products of 8 isolates were DNA sequenced and created phylogenetic tree. These 8 isolates were found to be very close to each others and radish isolate of CMV (Gene Bank No: LC066508) from Turkey by phylogenetic tree.
