Effect of argan oil on liver and kidney mitochondrial oxidative stress and respiratory enzymes in acrylamide given rats

Acrylamide (AA) is an important chemical that pose a risk to public health. Argan oil (AR) is rich in antioxidants and phenolic compounds. We aimed to study the protective effect of AR on mitochondrial oxidative stress and mitochondrial functions that may occur with AA in rat liver and kidney. For this purpose, Sprague Dawley rats were given with AA (50 mg / kg i.p., 3 days a week), AR (6 ml / kg o.p, per day) and AA together with AR for 30 days. Cytosolic enzymes glutathione s-transferase (GST), glucose-6P-dehydrogenase (G6PD) enzyme activities decreaded but myeloperoxidase (MPO) and total nitric oxide (NOx) levels increased in the AA treated rats. By the effect of AA, mitochondrial reduced glutathione (GSH) level and mitochondrial antioxidant enzyme activities decreased. In contrast, significant increases were seen in mitochondrial lipid peroxidation (LP) and protein carbonyl (PC) levels. In addition, significant decreases were observed in the activities of electron transport chain (ETC) and the tricarboxylic acid (TCA) cycle enzymes and also ATP and mitochondrial metabolic function (MTT) levels decreased by the effects of AA. AR administration together with AA improved significantly oxidative stress and mitochondrial function parameters caused by AA and most parameters were completely normalized. Findings show that AR is effective in improving mitochondrial oxidative stress and deteriorating mitochondrial functions in rat liver and kidney tissue with AA effect.
