SUMMARYDETECTION of mRNA PRODUCTS WHICH ARE PRODUCTS OF CHIMERIC GENESOCCURED BY TRANSLOCATION BETWEEN HOX AND NUP98 GENES INPATIENTS DIAGNOSED AS HAVING ACUTE/CHRONIC LEUKEMIA (AML,CML,)and MYELODISPLASTIC SYNDROME (MDS)Gizem AYNAAs a results of chromosomal translocations, NUP98 and various partner genes wereshown to form fusion products in human leukemias. Bone marrow samples of leukemia,myelodysplastic sendrome (MDS), and myeloproliferative disease (MPD) patients , sent toour laboratuary from the Dokuz Eylul University hospital and other hospitals in zmir, wereused in this study. The presence of NUP98- HOX (HOXA9, HOXA11, HOXA13, HOXC11,HOXC13, HOXD11, HOXD13) fusion products as well as NUP98-DDX10 fusion productswere investigated. Cellular total RNA was extracted from the bone marrow mononuclear cellsand complementer DNA reaction was set up to obtain double strand DNA. The ß-actin geneproduct was used as internal control. The cDNA products were amplified by PCR using 17different primer sets selected from medical literature.The ß-actin internal control was foundpositive in all reactions.In conclusion, NUP98-HOX fusion gene products were found to be negative in 34patients tested while the NUP98-DDX10 products were negative in the total of 30 patientstested.Key Words: Leukemia, translocation, NUP98, HOX, DDX10, chimeric gene, RT-PCR.
