ABSTRACT THE INVESTIGATION OF ENDOTHELIN 1 AND ADDUCIN GENE POLYMORPHISM IN PATIENTS WITH ACUTE MYOCARDIAL INFARCTION Coronary heart disease (CHD) leads to the first cause of morbidity and also mortality in the developed countries and as well as in our country. Endothelin 1 formed by vascular smooth muscle and showing paracrine and autocrine effects is an polypeptide with 21 amino acid. Adducin a ubiquitous cytoskeletal protein and can modulate a variety of cell functions, such as ion transportation by promoting the binding of actin spectrin. In this study, the presence of relationship of acute myocardial infarction with endothelin1 and adducin gene polymorphism was investigated in patients with acute myocardial infarction diagnosed at the Department of Emergency Medicine. The study group consisted of 50 patients admitted to and diagnosed at the Department of Emergency Medicine, Firat University Hospital and the control group consisted of 50 healthy person were included in the study. 2 or 3 cc blood taken from the members of the study group and put into the tubes with EDTA underwent DNA extraction at the Laboratory of Medical Genetics. Mutation in gene region were investigated by multiple PCR method. The mean age of the patient group was 60,46?13,81, the mean age of the control group was 59,30?12,4 (p> 0.05). Of the patiens with AMI was 41 males (82%), 9 females (18%), of the control group was 34(68%) males and 16 females (32%). In the patient group, mutan phenotype frequencies for endothelin 1 Lys198Asn, endothelin 1 rs10478694 and adducin Gly460Trp were found %18, 12% and 0%, respectively. In the control group, these were found %10, 8% and 6%, respectively. The incidence of Endothelin 1 and adducin genes in each of three genotypes was not significant (p> 0.05). As a result, it was shown that known risk factors play independent role from endothelin 1 and adducin for the etiopathogenesis of AMI, also endothelin 1 and adducin were not a risk factor for CAD.
