Abstract (EN)

The genus Alchemilla (Rosaceae) is represented by 78 species in Turkey, 26 of which are endemic Some species of Alchemilla are used in folk medicine for coughs, asthma, bronchitis, and wound healing. Phenolic and terpenic compounds have been isolated from Alchemilla species up to date. No isolation study was found on Alchemilla pseudocartalinica Juz., one of the species growing naturally in our country and recorded to be used as a diuretic and tonic against diarrhea in the Eastern Anatolia Region in ethnobotanical studies. Within the scope of the thesis, it was aimed to investigate various biological activities of A. pseudocartalinica and to elucidate the phytochemical content of the active extract by chromatographic techniques. The 80% methanol extracts were obtained from the aerial parts and roots of A. pesudocartalinica species, and sub-extracts were obtained from the crude extracts by liquid-liquid extraction method. Acetylcholinesterase, butyrylcholinesterase, tyrosinase, and α-glucosidase inhibitory activities of n-hexane, dichloromethane, n-butanol, and water sub-extracts were investigated by in vitro studies. In addition, the phosphomolybdenum-reducing antioxidant capacity (PRAP), metal chelating capacity, and iron-reducing antioxidant capacity (FRAP) of all extracts were measured. The water sub-extract with the highest α-glucosidase inhibitory activity was fractionated using column chromatography methods and the activity of the obtained fractions was investigated in the same test system. The structure of the major substance in the most active fraction was elucidated as quercetin 7-O-β-glucopyranosyl-3-O-β-glucuronopyranoside. In addition, LC-MS/MS method was used to elucidate the chemical profile of the extracts. Elagic acid (38,42 µg/g) in the aerial part methanol extract and catechin (185.30 µg/g) in the root methanol extract were found to be the compounds with the highest content.
