Abstract (EN)
Allogeneic Hematopoietic Cell Transplantation (AHCT) is an important and potentially curative treatment option for many malignant or non-malignant hematologic diseases. Worldwide it is known that peripheral blood is mostly used as stem cell source for AHCT. In our study, datas of 136 healthy donors who applied between 2013 and 2017 were retrospectively evaluated. With all the donors only peripheral blood source have been used. Our study group involved 49 female ( 36%) and 87 male ( 64%). CD34+ cell counts were relatively higher in males' peripheral blood sample that was taken before and after apheresis, although it was statistically insignificant (p =0,260). The study group's median year was 39. Eventhough specifically from donors under 40 more cells were gathered than those over 40, it was found statistically insignificant (p = 0.065). The group's median wieght was 76,00 kg. The CD34+ cell counts that were collected before apheresis and then later from blood products of donors who were over 76 kg were higher, and it was statistically significant (in order of, p= 0.034, p= 0.024). It was more significant in female donors that higher weight associated with higher CD34+ cell counts. There were higher cell counts in the group with BMI> 24 than the group with BMI< 24 (p = 0.004). Higher CD34+ cell counts were found in donors whose leucocyte counts were also higher than median levels on both basal hemogram tests and before apheresis tests (in order of, p=0.043,p=0.017). More CD34+ cells were harvested from donors who used Filgrastim as active substance compared to those who used Lenograstim (p=0.049). Apheresis were performed on 73 donors with Fresenius device and on 63 donors with Optia device. There were more CD34+ cells in blood products which harvested with Optia device (p= 0.038). On the donors who got apheresis only once, 59.7% of the time Optia device and 40.3% of the time Fresenius device were used (p=0,002). 12 of the donors (8.8%) showed hypocalcemia symptoms. There were no other side effects and/or catheter related complications. Our parametres in these studies were age, gender, weight, body-mass index, blood counts and peripheral blood CD34+ cell counts before G-CSF administration. The parametres that were found statistically significant correlate with other literature researches. In all the donors we included in our study aimed peripheral blood CD34+ cell levels before apheresis (˃10/μL ) were achieved and on all the donors apheresis were performed. Our study is the only one that compares Optia and Fresenius devices.
