Abstract (EN)
Wheat is an important agricultural crop cultivated in Turkey and all around the world. Although many weed species caouse significant problem in wheat cultivation, herbicide application constitute the most effective management method. Despite herbicide application is an effective method, resistance problem arises in weeds due to continuous applications. The aim of this study was to determine resistance of different Sinapis arvensis populations against tribenuron methly and 2,4- D amine herbicides by bioassay and molecular methods. On this purpose wild mustard seeds were collected from 310 different wheat fields located in Amaysa, Tokat, Çorum and Yozgat provincces. When seeds subjected to registered dose of tribenuron methyl in bioassay, the survival of thirteen populations were observed. Populations that survived in the registered dose application were subjected to dose-effect studies, herbicide application was applied up to 16 times and their symptoms were recorded. For cross resistance studies populations were subjected to 2,4 amine applications and cross resitance was not found in thirteen populations. Point mutation causing ALS resistance in wild mustard was determined by PCR test of B domain in same populations. As a result of the PCR tests, DNA fragment analysis was performed and it was determined that the Trp-574 amino acid was transformed into Leucine amininoacid as a result of the point mutation. No mutation was detected in other populations. In this study the resistance resulting from the point mutation in the amino acid Trp-574 in wild mustard was determined molecularly for the first time in Turkey.
