Investigation of the protective effect of linoleic acid in the ß-amyloid-induced in vitro alzheimer's disease model

Abstract (EN)

This study was carried out to investigate the effects of Linoleik Asit (LA), known as Omega-6, on apoptotic pathways and antioxidant enzymes in the β-amiloid (Aβ) induced in vitro Alzheimer's Disease (AD) model. In the study, 3 groups were formed: the control group consisting of U87 cell lines, the Aβ group in which the in vitro model of AD was created containing Aβ induced U87 cell lines and the Aβ+LA group obtained by adding LA to the Aβ group. In the study groups, caspase-3 representing apoptotic pathways, and superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) enzyme levels and Total Antioxidant Status (TAS) and Total Oxidant Status (TOS) results were analyzed to evaluate the antioxidant status. Compared with the control group, in the U87 cell line exposed to Aβ, it was determined that SOD, CAT, GPx, TAS levels decreased significantly; caspase-3 and TOS levels increased significantly. However, it was found that applying LA to Aβ-U87 cell line significantly increased SOD, CAT, GPx, TAS levels and decreased caspase-3 and TOS levels. As a result, it was revealed that LA decreases the neurodegeneration due to the apoptosis and increases the antioxidant activity in a manner that supports it. Considering these effects of LA, it has been evaluated that this powerful antioxidant in AD may have the potential to be used in protective or alternative treatment, and this should be supported by further studies.
