Lipases are an enzyme that catalyzes the hydrolysis of lipids. Lipases are a subclass of the esterases. Lipases perform essential roles in the digestion, transport and processing of dietary lipids ( triglycerides, fats, oils ) in organisms.Lipases serve important roles being exploited as cheap and versatile catalysts to degrade lipids in more modern applications. In the present work pistachios seed is choosen as an alternative source for purification and characterization of lipase. By purification of lipase from pistachios seed procure rem, deoiling of pistachios seeds protein procure ammonium sulfate precipitation purify by dialysis gel filtration chromatography methods. Protein concentration was determined by the Bradford method using Sephadex G-100 gel filtration column chromatography. In the and specific activity has been calculated as purification coefficient of 1.97 fold. Lipase activity was highest at PH 8.8 and at 45°C . Also stable activity showed at PH 8.4 and at 45°C. Pistachios seed's lipase stability is been tested for 120 hours and at the end of the test it has showed thah it's stability is protected at 45°C. Lipase substrate specificity has been calculated Vmax and Km. Their Vmax 15,60 U/dk.mg and Km 0,078 Mm enzyme, respectively.
