Abstract (EN)
In this thesis, the enzyme activities of plant extracts of Abies, Pinus and Juniperus species against α-amylase, α-glucosidase and lipase enzymes were investigated. In addition, the possible binding patterns of 25 phenolic compounds in these extracts to enzymes were determined by in silico studies. Juniperus oxycedrus subsp. oxycedrus leaf part of methanol (IC50= 8µg/mL) extract and Juniperus oxycedrus subsp. oxycedrus the methanol (IC50= 6µg/mL) extract of the fruit part of the oxycedrus species inhibited the α-glucosidase enzyme significantly. Juniperus oxycedrus was determined that the methanol extract (IC50= 74µg/mL) of the leaf part of the Juniperus oxycedrus also had a high inhibitory effect on the α-amylase enzyme. It was determined that the water extract of the leaf part of the Pinus brutia Ten species had an inhibitory effect against the lipase enzyme with an IC50 value of 45µg/mL. As a result of the in silico study, the binding energy of the hesperidin molecule against the enzymes was found as -159,952 MolDock Scores -160.057 MolDock Scores and -156.552 MolDock Scores for α-amylase, α-glucosidase and lipase enzymes, respectively. It is seen that hydrogen bonding, hydrophobic and van der Waals interactions make important contributions to the interaction of molecules with enzymes.
