Investigation of the effect of snps in stat5a gene on milk yield in Anatolian buffaloes

STAT5 genes play an important role in mammary gland development, milk protein synthesis, mammary glandular cell proliferation and regulation of milk-protein gene expression, mediating PRL and GH signals, fertilization and embryonic development. The STAT5A gene, which has important roles in mammary gland formation and lactation, is among the candidate genes to be investigated, especially in milk-producing animals. In this study, hairs were taken from 192 Anatolian buffaloes in Afyonkarahisar region and DNA Sequence Analysis process was applied. By adding milk yields to the findings obtained, the relationship between polymorphic regions and milk yield was tried to be determined. In the study, the 1720 bp long area between the 6-9 exons of the STAT5A gene was examined. As a result of DNA Sequence Analysis, a total of 13 polymorphisms were detected, 5 in the 7th intron, 7 in the 8th intron and 1 in the 8th exon. By adding milk yields, association analysis was made with the help of statistical program and it was determined that there is a statistically significant relationship between SNP12237 C>T polymorphism in the 8th exon and milk yield. As a result, SNP12237 can be used to increase milk yield in buffalo breeding studies. It would be beneficial to investigate the STAT5A gene more extensively for breeding buffaloes.
