Isolation, characterization and investigation of some probiotic and starter features of lactic acid bacteria from human breast milk

Abstract (EN)

Live microorganisms showing positive effects on host health are called probiotics. Besides milk is an important food source for newborn babies contains lactic acid bacteria showing probiotics characteristics. In this study, Lactic acid bacteria isolated from breast milk. After isolation it has made the characterization of isolates by some phenotypic method. Then the identification of isolated bacteria were done with API 20 Strep system. End of this identification two Streptococcus salivarius and one Lactococcus lactis subsp. cremoris strain are obtained and continued with these strains in this work. Determined these three strains are examined for starter feature. As these isolates have shown growth in salt concentration between 4% and 5%. At last 48 hours, pH concentrations are measured and acid production rate are monitored between pH 5.40 and pH 6.93 for these three strains. Also exopolysaccharide production of these strains was determined and S. salivarius (M14-365) was observed the highest rate (8.71 g/L). EPS production of L. lactis subsp cremoris (M4-157) and S. salivarius (M4-11) strains were determined as 4.94 g/L and 5.49 g/L respectively. It was determined that all strains were resistance against nisin. Some probiotic features of these strains were investigated. And they were tested with antibiotics (ampicillin, gentamicin, kanamycin, erythromycin and penicillin G ) was found to be sensitive to this antibiotics except three strain kanamycin antibiotic. Besides antimicrobial activity on various human and fish pathogens of these strains was investigated. Only S. salivarius strains (M4-11 and M14-365) showed antimicrobial activity against P. aeruginosa pathogen of human.
