Protective effects of resveratrol aganist arsenic toxicity on testis

PurposeIn this study we aimed to investigate protective effects of resveratrol on arsenic induced toxicity, apoptosis and oxidative stress on testis.Material-MethodIn this study, adult male Wistar rats (200-300 gr) were used. Rats were divided in five groups. Group I: Conrol group: Non- arsenic group, rats were fed ad libitum (n=7). Group II: Alcohol group: Because of resveratrol was soluable in alcohol, to see the effects rats were given alcohol by intragastric rout (n=7). Group III: Arsenic group: Rats were given 5mg/kg/day arsenic by intragastric route. Group VI: Arsenic+Resveratrol group: Rats were given 5mg/kg/day arsenic and 10mg/kg/day resveratrol by intragastric route (n=7). Group V: Arsenic+Resveratrol group: Rats were given 5mg/kg/day arsenic and 20mg/kg/day resveratrol by intragastric route (n=7). Experimental process was chosen 28 days for all groups. After the experimental process, oxidative stress marker malondialdehid (MDA), antioxidant enzyme glutathione (GSH) and TUNEL and Caspase 3 immunohistochemistry for apoptosis were examined in each group. Light microscobic and electron microscobic observations were used for ultrastructual changes. All data were analyzed by one-way ANOVA posthoc LSD test. p<0.05 was considered statistically significant.ResultsMDA levels in arsenic group showed a significant increase compare with control group. GSH levels were significantly lower than those control group. Light microscobic and electron microscobic investigations showed that on arsenic tissues, seminifer tubules showed vacuolization, atrophy, germ cell degeneraion and celluar debris on tubul lumens. After immunohistochemical investigations apopitotic cell death was increased in arsenic group compared with control group. Resveratrol decreased TUNEL(+) cells and caspase 3 immnunostaining showed no increased apoptosis on treated groups compared with arsenic group.ConclusionIn conclusion, the protective effects of resveratrol on arsenic induced toxicity on testis was demonstrated firstly in the present study. Resveratrol exhibits antiapopitotic effect via with its antioxidant properties.Key words: Arsenic, testis, resveratrol, apopitosis, oxidative stress
