Abstract (EN)

Age-related macular degeneration (AMD) is an important cause of vision loss, with an increase in average human life, especially with increased frequency of developed features. There is still no definitive treatment that prevents the progression of YBMD with age-type anti VEGF drug treatment. With the understanding that inflammation and oxidative stress play a role in the pathophysiology of AMD, the search for new treatments has accelerated. Acetyl L-Carnitine (ALKAR) is an agent known for many years and used as a supplement in various pathologies. In previous studies of ALKAR, many defaults have been shown for anti-inflammatory activity and antioxidant activity, and finally there is also anti-VEGF feature in in vivo effective cancer cells. We chose anti-VEGF and antioxidant in ALKAR's retinal pigment epithelium to show them. It was seen to create hypoxia model with 800 µM CoCl2 to form hypoxia model with 800 µM CoCl2. In the ALKAR dose study, it was observed that 10 µM was the effective dose. It was seen that the ALKAR group was statistically significantly increased (p <0.05). ALKAR performed with light microscopy was also shown to have a protective effect on the morphological evaluation of ARPE-19 cells against hypoxia. In addition, Bevakizumab reduced cell viability and ALKAR was shown to have a protective effect when ALKAR was used together. ALKAR was shown to show anti VEGF by reducing CXCR-4 and HIF-1α. Total Antioxidant Capacity (TAK) was examined and a statistically significant difference was found in the group given ALKAR (p> 0.05). The newly detected anti-VEGF feature of ALKAR, which has been used for a long time, suggests that hypoxia is beneficial in pathophysiology use of space retinal diseases, especially AMD.
