Investigation of dose and time depedent effects of pentoxifylline on in vitro sperm parameters of asthenozoospermia and normozoospermia patients

Abstract (EN)
INTRODUCTION AND AİM: In this study, we aimed to observe the in vitro effect of pentoxifylline on sperm motility and viability in terms of dose and duration in asthenozoospermia and normospermia patients and to find the most suitable concentration and processfor sperm. MATERIALS AND METHODS: In our study, semen samples from 40 patients (20 normospermia and 20 asthenozoospermia patients) who applied to the urology clinic were obtained. The samples were divided into control and experimental groups. Wash solution-distilled water mixture were added to the control group. We added 3,6; 4,8 and 5,6 mM pentoxifylline separately to experimental group samples. The control and experimental groups were incubated with these solutions for 10 minutes and 20 minutes. Makler counting chamber the sperm count was made by the square system. Numerical values were examined in three groups; progressivemotility (A), nonprogressivemotility (B) and nonmotility (C). Vitality staining was carried out and the viability of the sample was examined with the aid of a light microscope. RESULTS: As a result of light microscopic observations, asthenozoospermia and normospermia group were compared with each other in group A and statistically significant increase was found in both groups. It was observed that this increase was more effective at the dose of 5,4 mM and during 20 minutes of incubation. When group A motility was examined in the multiple comparison test, it was seen that this increase was statistically more significant in patients with asthenozoospermia. In both patient groups, a statistically significant increase was detected in the B group comparison results, and this increase was noticeable in 20 minutes. When the concentration subgroups were compared in asthenozoospermia patients, a statistically significant decrease was observed in group C, although it decreased in normospermia patients, but this decreasewas not statistically significant. There was no statistically significant difference in viability evaluations between the two groups of patients. CONCLUSION:The use of pentoxifylline in the asthenozoospermia and normospermia patients increased the number of sperm samples A and B statistically, where as the C group had conflicting results. This suggests that pentoxifylline activates nonmotility sperm.
